Experimental Process

Visual inspection of the algD promoter identified two sites matching IHF consensus sites in E.coli. EMSA confirmed binding of IHF to the algD promoter. The author identified that mutations in each of the sites resulted in reduction or loss of in vitro IHF binding and a three- to fourfold decrease in algD-cat expression.
