Experimental Process

lac-Z assays showed that ohr expression is repressed by OhrR. GUS reporter gene assay showed equivalent results. Visual sequence inspection identified two palindromes resembling the OhrR binding site. EMSA results showed direct binding of OhrR to both motifs. Site directed mutagenesis in conjunction with EMSA confirmed the importance of these motifs for the the binding by OhrR.
