Experimental Process

Visual inspection revealed five consensus covR binding sites upstream of covR. EMSA of this region confirmed CovR binding. DNase I footprinting localised the regions for CovR binding. Site-directed mutagenesis of thymine residues in the identified sites in conjunction with DNase I footprinting validated their identity as binding sequences. GUS reporter assays confirmed the role of CovR as an repressor.
