Experimental Process

lacZ assay results suggest that AraR represses the araABDLMNPQ-abfA operon. Site directed mutagenesis showed that mutations to the binding site increased expression of the araABDLMNPQ-abfA operon. in vitro transcription confirmed that AraR repressed the araABDLMNPQ-abfA operon. DNase I footprinting determined the binding sequence.
