Experimental Process

Quantitative real-time PCR showed that the level of hupA transcript was reduced in the crp mutant compared to the wild-type strain. Primer extension analysis performed using the wild-type and the crp mutant strains confirmed that the expression of hupA in V. vulnificus is under the positive control of CRP. EMSA confirmed that CRP activates hupA expression by directly binding to the hupA promoter. DNase I footprinting showed that CRP protected a region containing a consensus CRP binding site.
