Experimental Process

vvsA-luxAB reporter fusion showed that vvsA expression repressed by Fur in the presence of iron. Visual inspection of the vvsA promoter identified a Fur box with only two nucleotide differences from the consensus Fur binding site. EMSA showed that Fur bound specifically to the vvsA promoter. DNase I footprinting identified that Fur binding region contained the putative Fur box.
