Experimental Process

A putative CcpA binding site was identified in the tst promoter region by screening S. aureus N315 with the CcpA consensus sequence. Multiple sequence alignment confirmed that this sequence was conserved in tst promoters of the strains N315 and RF122. Luciferase reporter assays identified that glucose-dependent repression of tst transcription required CcpA.
