Experimental Process

A rpoN knockout mutant strain of P. aruginosa PA01UNSW was created to test the effect of RpoN on the expression of rhlI. A lacZ fusion assay determined that RpoN positively regulates rhlI transcription, demonstrating its role as an activator of rhIl. Alignment of promoter of genes known to be regulated by RpoN identified an RpoN consensus sequence (YTGGCACGrNNNTTGCW).
