Experimental Process

Microarray analysis identified the PsrA regulon. The microarray data was confirmed by qRT-PCR for select genes. fadBA5-lacZ expression showed that fadBA5 expression was dependent on presence of PsrA and long-chain fatty acids. EMSA verified that PsrA binds specifically to the fadBA5 promoter. Visual inspection identified two PsrA consensus sequences in the fadBA5 promoter. Consensus search and binding site alignment identified additional PsrA binding sites in the genes contributing to Fad.
