Experimental Process

LacZ reporter assays in wild-type, fleQ and rpoN mutant strains confirmed FleQ and RpoN dependent regulation of the target promoters. EMSAs showed FleQ specifically binds to sequences within its target promoters. DNase I footprinting was used to identify FleQ binding sites. Site-directed mutagenesis of the FleQ binding site in the flhA promoter confirmed direct regulation by FleQ.
