Experimental Process

5′ RACE method was used to identify the XCC2731 transcription start site. A predicted Clp-binding site TGAGCN6TCACA, with 7/10 matching the consensus sequence, was located at −87/−102 relative to the XCC2731 TIS. XCC2731–lacZ transcriptional fusion constructs were used to map the upstream region required for expression of the XCC2731 gene. EMSAs demonstrated that Clp binds its target promoter.
