Experimental Process

Transcriptional profiles of PAO1 and the isogenic cueR deletion mutant were constructed to identify the role played by CueR in the response to copper. Using the E. coli CueR binding motif, the researchers searched for potential CueR binding sites in the P. aeruginosa genome. Promoter-lacZ transcriptional fusions for the five putative promoter targets of CueR were used to validate the microarray results. The results of these assays showed that all five promoters were activated by the addition of copper. EMSAs were done to verify the direct regulation by CueR of its target promoters.
