Experimental Process

fur::luxAB transcriptional fusion showed that the Fur protein activates its own expression, and the extent of its activation is elevated in the presence of iron chelator added to the growth medium. Specific binding of Fur to the fur promoter was confirmed by performing DNAse I footprinting assays. Mutagenesis of the putative Fur box and a fur(mutant)::luxAB transcriptional fusion confirmed that the interaction between Fur and the protected DNA sequence is essential for autoregulation of the fur gene.
