Experimental Process

Putative PerR binding sites were identified in the S. aureus based on homology with the putative site upstream of katA. To determine whether these candidate PerR binding sites were located in the promoter region of the genes, transcriptional start point was determined. In each case, the PerR binding site was located in the promoter region close to the −35 and −10 elements. LacZ reporter fusions were used to confirm PerR-regulated expression of the identified genes.
