Experimental Process

Putative Fur-binding sites in the genome of C. crescentus were detected by using a position weight matrix approach. EMSA validated that Fur binds to its target promoter region. LacZ reporter assays validated Fur-mediated expression regulation. Site-directed mutagenesis of Fur-binding sites confirmed specific binding.
