Experimental Process

Using DNAse I footprinting the Fur specific operator sequences were identified and this also confirmed the binding of Ec-fur to primary and secondary sites. To examine the influence of iron and Fur on the expression of fbpA a gene fusion between the promoter regions of fbpA and chloramphenicol acetyltransferase (CAT) was constructed and PCR amplified to test gene expression.
