Experimental Process

Genome-wide DNA microarray hybridization was done to identify Zur-regulated genes. gusA transcriptional fusion assays were performed to confirm Zur-mediated expression of these genes. EMSAs were done to confirm that Zur directly regulates its target genes. To further clarify whether Zur directly represses the transcription, in vitro transcription assays were performed. DNaseI footprinting analysis was employed to locate the Zur-binding sites within the promoter regions.
