Experimental Process

A beta-gal assay was used to show that PhhR activates phhA expression. A PSSM search was used with the motifs of IHF (another NtrC TF) to identify a proximal and distal site. A beta-gal assay was used to determine that the distal site is necessary for phhA expression. EMSA and DNAse footprinting were used to show binding.
